EGCG decreases myocardial infarction in both I/R and MIRI rats through reducing intracellular Ca2+ and increasing TnT levels in cardiomyocytes

BACKGROUND Myocardial ischemia/reperfusion injury (MIRI) usually induces serious health problems. OBJECTIVES This study attempted to explore protective effects of (-)-epigallocatechin-3-gallate (EGCG) on MIRI and the associated mechanism. MATERIAL AND METHODS Ischemia/reperfusion an isolated rat heart (I/R model) model were used in this study. infarction was measured with staining 2,3,5-triphenyltetrazolium chloride (TTC). Ca2+ troponin T (TnT) concentrations coronary perfusion fluid evaluated using chromatometry method. concentration cardiomyocytes determined detecting fluorescence intensity. The ultrastructure observed transmission electron microscopy (TEM). β-nicotinamide adenine dinucleotide (NAD+) also determined. RESULTS EGCG (I/R+EGCG) significantly reduced myocardial size compared I/R rats (p < 0.05), remarkably increased decreased TnT as well markedly intracellular promoted NAD+ 0.05). It obviously maintained mitochondrial structure improved rats. Lonidamine (LND) treatment (I/R+EGCG+LND group) blocked I/R+EGCG group (MIRI+EGCG) 0.05) enhanced pulmonary artery that CONCLUSIONS both models by reducing concentration, increasing promoting improving cardiomyocytes.
